Carbohydrate Name

Ssp-4 (amastigote surface glycoprotein)

Carbohydrate Class

Miscellaneous

Source Microbe

Trypanosoma cruzi amastigotes

Basic Structure

Ssp-4 is an amastigote-specific 84 KDa surface component defined by monoclonal antibody (MAb) 2C2, that was shown to be linked to the membrane by a GPI anchor. The epitope defined by MAb 2C2 is homogeneously distributed over the amastigote surface throughout the intracellular life cycle until it is gradually shed through a phospholipase-dependent mechanism during transformation into trypomastigote

Proposed functions

N/A

Antigenic Nature used to produce antibodies

mixtures of heat-inactivated intracellular and extracellular amastigotes

Carrier Name

nil

Conjugation Method

nil

Antibodies

Mab 1D9

Antibody type and class

IgG3

Assay System

ELISA, immunoblotting, immuno-electron microscopy and confocal immunofluorescence

Cross-reactivity

This Mab cross-reacts with membrane of amastigotes from 569, 588, 573, 587 and SC2005 isolates, similar to the G isolate. It also reacts with the CL isolate

Proposed epitopes

N/A

Proposed Utility

This Mab reacted with cytoplasmic membrane-bound compartments of the amastigotes. This Mab specifically inhibited cell invasion by G and 588, but not CL strains
